Defining how CDK7 kinase controls RNA polymerase II function at gene 3'-ends
CDK7 regulates RNA polymerase II (RNAPII) at multiple stages of the transcription cycle, yet the mechanisms governing 3' end termination remain poorly understood. My project investigates how CDK7 inhibition disrupts transcription termination, leading to widespread 3'-end readthrough, using integrated multi-omics encompassing nascent transcriptomics, quantitative proteomics, and phospho-proteomics. Additionally, I am complementing these approaches with single-molecule TIRF microscopy (RIFT) that enables biophysical interrogation of CDK7-related transcription dynamics in a reconstituted system.
